Who's got it, and playing it?
I got into the beta the other day after months of waiting. Never played the first one. Consider myself to be an adept game player but the learning curve seems fairly big. In around 2 hours of game time I've failed to kill a 'Hero' (either human player or bot) but died numerous (read: many) times. Lol.
Need tips!

Play with bots until you learn the mechanics of the game. If you want to play in MM stick with heroes you know and subscribe to DotaCinema and Purge for hero introductions, guides and how-tos. You can go to the Dota 2 subreddit plenty info there and join their newbie channel and play inhouse matches.

Some good advice, vladTGS. Thank you.
After quite a lot of reading/watching on YouTube and Twitch as well as playing bot matches, I decided to give online another go. The good news is that not only did my team win but I managed to get myself 8 kills and 1 assist, with only 3 deaths. A personal best.
Two characters I found useful to learn with are Lich and Venomancer. Each time I learn more about the game, it just seems to get better.

It's good you started out with support heroes, that way you can work your way up the ladder more easily to semi carries, hard carries etc. learning to last hit, deny, position, initiate and gank along the way. When you random( I do that sometimes just for the fun of it) and you don't know how to build your hero just use this site : http://www.dota2alttab.com/ . You don't have to buy exactly those items or skill the hero that exact way, each game is different so you have to adapt, it's just to get a general ideea.

I'm up on last hit and denies, although my timing is off a little at some points. Haven't went anywhere near ganking yet, tend to stay in lane to learn the basics properly.

I think I've now officially elevated myself to matchmaking instead of playing just with bots (as much as that will irritate some online players, Lol). Capable enough to get some kills, even though there's still loads and loads (and loads...) to learn.
Also started taking the advice given by Icefrog to Gabe Newell to always play random. Better chance to learn the characters, although painfully; at times.
The game is turning into a little diamond. Can't wait to see the community explode when it hits full release.

Starting to have a great time with Dota 2.
Current play time is around 19 hours but I've only just started to get more kills than I have deaths in most matches (though it's not consistent). Playing random to learn more about the Heroes and sticking to my lane unless someone directs elsewhere; such as a push - which can be awesome carnage when working as a team.
My focus is currently on a decent farm/support, last hits/denying and harassing without going too far towards the 'Don't feed, you noob' status.

If I read it correctly, Dota 2 now has a proper page on Steam with a 'coming soon' in the release date. Might be getting close!
Been frustrated with the game the last few nights, though. Two things becoming more common: matchmaking taking multiple attempts to get into a game and people dropping out shortly after 'first blood'. I played against 5 enemy Heroes on my own last night...

Great stuff, it's easily one of the best games I've played this year. And upon release, the fact it's going to be free 2 play is amazing.
Word of advice is to start out by playing bots on easy and get a feel for the game, how it works etc. Jumping right into matchmaking may feel 'out of your depth'. Lots of resources on the web to help understand it, too. Get reading, Lol.
I'm really starting to get a good grasp of how to play well. Only 35 hours in so still lots and lots to learn but nearly every game is enjoyable and definitely educational.
